Inventing Challenge

If you miss that "all important" phone call for the challenge does that mean you can't do the challenge? Or will they call again?

Are you referring to the opportunity to make a Simbot?  How did you miss it exactly?  If you didn't accept it or cancelled it, you should get it again eventually.  It might take some time or it might happen quickly.  I can't say for sure.

You'll get the opportunity pop-up even if you don't answer the phone.

When your Sim's phone rings, if it's an opportunity calling it will pop up on the screen even if your Sim doesn't answer the phone.  You'll never lose opportunities that way.  If it rings and nothing pops up, then it was another Sim calling to chat.  No major loss there.
